Pentosan polysulfate has one of the strongest evidence bases of any veterinary joint therapy. It is approved for use in dogs in multiple countries (Australia, UK, EU) and Adequan (a related polysulfated glycosaminoglycan) is FDA-approved in the US. Published veterinary research demonstrates inhibition of cartilage-degrading enzymes (metalloproteinases), stimulation of cartilage matrix synthesis (proteoglycan and hyaluronic acid production), improved synovial fluid quality and joint lubrication, anti-inflammatory effects within the joint, and improved clinical lameness scores in controlled veterinary trials.

🩺 Administration: PPS is administered by subcutaneous or intramuscular injection. Adequan (US) is given intramuscularly. Cartrophen Vet (Australia/UK) is given subcutaneously. The typical loading protocol is one injection per week for 4 weeks, followed by maintenance injections every 4-6 weeks. This is a veterinary prescription medication administered by or under the direction of a licensed veterinarian. Not available OTC.
Side Effects & Safety
PPS/Adequan has a well-documented safety profile from decades of veterinary use. It is an anticoagulant (blood thinner) at therapeutic doses, so bleeding risk is the primary safety concern. Side effects include potential prolonged bleeding time (use with caution pre-surgery), injection site pain or swelling (usually mild), rare GI upset (diarrhea, vomiting), and very rare allergic reactions. Contraindicated in dogs with bleeding disorders, those on anticoagulant medications, or dogs scheduled for surgery within 2 weeks. Liver and kidney function should be normal before starting.
⚠️ Tumor Caution: Peptides promoting tissue repair or growth could theoretically accelerate existing tumors. Vet screening recommended before starting, especially for seniors and cancer-prone breeds.
